Researchers have created the GoT-Multi, a single-cell multi-omics method that allows for concurrent analysis of genetic and transcriptional heterogeneity in fresh and FFPE tissues. This innovative technique integrates multiplexed genotyping with single-cell RNA sequencing, facilitating the study of somatic mutations and gene expression in therapy-resistant cancers. Applied to chronic lymphocytic leukemia tumors, it revealed complex tumor ecosystems with overlapping transcriptional profiles among distinct genetic subclones, thereby offering insights into tumor heterogeneity and resistance mechanisms, which may aid clinical diagnostics.
